UV-VISIBLE DATA COLLECTION FOR Fe(II) AND TOTAL Fe

  1. Install the Routine Sample Analyzer (RSA) into the Cary 5 Spectrophotometer

 

  1. Start the Cary V computer program by clicking on the CaryWinUV icon on the desktop

 

  1. Select the “Concentration” sub program.

 

  1. Turn on the Spectrophotometer and allow to warm up for 20 minutes.

 

  1. Select “File”, “Open Method”, then choose YOUR folder (user defined)

 

  1. Select “Set Up”, followed by:

 

*         “Choose standards” to set the number of standards -- note that sometimes blanks don’t count as standards.

 

*           “Choose samples” to set the number of samples

 

  1. Select “File”, “Save Method As”, then save into YOUR folder on Drive C

 

  1. Select “Zero It

 

  1. Select “Rinse” (which appears on a pop-up window) one time to drain the “Sampling Tube”
     line, then immerse the “Sampling Tube” into Dionized Water and rinse again at least 5 times, draining the tube between each rinse

 

  1. Immerse the “Sampling Tube” into Deionized Water, then select “Fill and Read” as your primary reading.  (The reading for water should be ZERO, or at least close to Zero).

 

  1. Select “Start”, followed by “Save as”.  This time save data in a file named “YOUR data” (user defined) in YOUR folder on Drive D

 

  1. Repeat steps 10 and 11 for each sample.

 

HINTS:

 

  1. Between each reading of different samples, Rinse the “Sampling Tube” by draining first, then refilling with Deionized Water to remove any trace left by the old sample.

 

  1. RINSE the “Sampling Tube” TWICE with the sample solution before taking the final reading in order to minimize errors.  Make sure that the “Sampling Tube” is fully immersed into solution

 

VERY IMPORTANT:  Upon completion of all samples, RINSE the “Sampling Tube” with Deionized Water for AT LEAST 20 times before turning the equipment off.

 

15. Perform calculations using Fe analysis data processing spreadsheet.
